SiR Action cell imaging
The imaging of SiR action is best done using standard Cy5 settings. After labeling, live cells can be immediately imaged without the need for washing steps. Optionally, a simple washing step includes replacing the labeled solution with fresh culture medium without probes, which typically improves the signal-to-noise ratio. If time elapses for imaging, it is recommended to keep the probe concentration equal to or lower than 100 nM throughout the experiment to obtain a constant signal and avoid probe interference with actin dynamics (reducing cell proliferation).
